Karam do you have another carb on hand that you can swap out. Few years back I restored a constellation and exact same story as yours. Tried every thing and checked every thing over and over again. Nothing. Then I substituted the Amal carb with a Mikuni and fired right up. Upon investigation it was found that the Amal carb I had on was ment for a 2 stroke. May want to try that out.
