Last time I used tyre foam in a bike tyre, it took a good three miles of riding for it to fully seal so there is definatley a dynamic process going on.
One of the first puncture sealants "gloops" out there was called linseal. This was designed for plant and agricultural vehicles which are mostly tubed. I've used it in motorcycle tyres before. By definition, I have no idea if it ever prevented a puncture. I certainly never got one with it.
