If yours is the Watsonian-Squire Dell'Orto conversion the carb is sealed into the alloy flange adapter with a couple of O rings. If for any reason these have failed you could be getting an air leak. Twisting the throttle will pass some petrol through the accelerator pump if it's a PHF32 carb, but you could still have a blocked pilot jet (the top one here, courtesy of ebay!)

Try this. Remove spark plug, pour a small amount of petrol into the cylinder (1/4 teaspoon) then quickly replace plug and attempt to start.
If it starts, or even coughs, you know you have a fuel delivery problem.
Unless you have a spark tester of some sort you don't know if the plug is sparking inside the cylinder. Compression can prevent a spark if there's a problem.
Anyway, try that and report back.
