Not sure what you mean by the nipple? The clip on the float should be at the top, not the bottom. Try it the other way up!

sorry I mean the small nipple that sticks out of the very bottom of he float bowl itself, not the float.
I am thinking it could be clogged up which is raising the needle up too high.
I notice it will start when I fill the float with fuel but as soon as I open the strangler to let air in the engine cuts out.
any more advice?

Well if it runs until you open the strangler, fuel is getting through, yes? What the strangler does is richen the mixture by restricting air supply, therefore when you open it it weakens the mixture which suggests that perhaps your throttle stop is set too high or the mixture screw is too far out. Throttle valve closed should be almost closed, say just 15thou open. Mixture screw should be approx 1.25 to 1.5 turns open. The engine will need to be well warm before you can start to gradually open the strangler, by the way.

Sorry for such a stupid question, but I only see 2 screws on the carb. One is horizontal and one below that is at an angle.
Are these the idle and mix screws?

Horizontal screw is mixture, the one at an angle lifts the throttle slide off it's stop. If you haven't already cleaned the carb up I think I'd take it off and clean it thoroughly.

I have loosened the acres by 1.5 turns but then the bike start the vibration causes be screws to loosen even further.
this can't be normal?

There should be a small spring under the mixture screw head to hold it in place, even if it's not there it shouldn't be loosening like that, it may be heavily worn. Replacements available from our Hosts. If you haven't already got one, get a Parts Book for this model, it will save a lot of time (yours and mine!). Available from our Hosts as a reprint or lots of genuine ones on Ebay. Does this mean you have it running consistently (not necessarily well!)?
