Can do. I would imagine, don't hold me to it, it depends on the distance between engine sprocket and swinging arm pivot.
The bigger that, the bigger the difference in slack.

I only know of the BMW G series that has its engine sprocket in line with the swinging arm, so there is no stretch, only wear.

So here's a quasi-official servicing lesson from an RE dealer in India -- quite a large one, by the looks of it -- where the chain tension is measured with the bike on its centre stand. The instructor mentions tightening with weight on the wheel, however, and there's a lot of noise, so it's not 100% clear...
